The Pavlovian model of tolerance holds that each administration of a drug constitutes a conditioning trial in which the drug is an unconditioned stimulus (UCS) (Pavlov, 1927). Environmental cues predicting the drug's homeostatic disruption become associated with the UCS as conditioned stimuli (CS). Over repeated pairings, CSs become able to initiate regulatory countermeasures (conditioned response, CR) in advance of the disruption, resulting in the decreased physiological responding known as drug tolerance. (Woods & Ramsay, 2000). Once tolerance has developed, cessation of drug use is generally associated with withdrawal. Partial reinforcement is a schedule in which the UCS is paired with the CS on fewer than 100% of the trials. Under these conditions development of the CS+UCS association, and thus the CR, is reduced. These experiments investigated the effects of partial reinforcement on the development of morphine tolerance and severity of withdrawal. Experiment 1 sought to replicate the established finding that partial reinforcement reduces the development of tolerance, and additionally to demonstrate that this results in less severe withdrawal. Experiment 2 examined whether partial reinforcement would have the same effect if subjects had a history of morphine, and Experiment 3 investigated whether the effect would hold if withdrawal were precipitated in a novel environment. Results of Experiment 1 showed that partially reinforced rats displayed significantly less analgesic tolerance, and less mastication during withdrawal, than those who were continuously reinforced. Experiment 2 found that partially reinforced subjects with a history displayed less tolerance than those continuously reinforced, yet these differences were not found with no history. On measures of withdrawal, there were no differences between groups in either history. Experiment 3 found no significant differences between partial and continuous reinforcement on tolerance or withdrawal mastication, but found that partially reinforced subjects experienced less weight change than continuous when withdrawing in the drug environment. These experiments provide the first demonstration that a manipulation of classical conditioning can affect not only the development of tolerance, but also subsequent measures of withdrawal. These results would be difficult to explain using a strictly pharmacological explanation, and so they provide strong evidence for the conditioning model of drug tolerance.
